背景介绍：Apoptosis is a cell death process that removes toxic and/or useless cells during mammalian development. The apoptotic process is accompanied by shrinkage and fragmentation of the cells and nuclei and degradation of the chromosomal DNA into nucleosomal units. DNA fragmentation factor (DFF) is a heterodimeric protein of 40-kD (DFFB) and 45-kD (DFFA) subunits. DFFA is the substrate for caspase-3 and triggers DNA fragmentation during apoptosis. DFF becomes activated when DFFA is cleaved by caspase-3. The cleaved fragments of DFFA dissociate from DFFB, the active component of DFF. DFFB has been found to trigger both DNA fragmentation and chromatin condensation during apoptosis. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene but the biological validity of these variants has not been determined.

The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

1、染色过强？

原 因：抗体浓度过高或孵育时间过长

解决方法：降低抗体滴度、抗体孵育时间：室温1小时或4度过夜

原 因：孵育温度过高超过37度

解决方法：一般在室温20-28度

原 因：DAB显色时间过长或浓度过高

解决方法：显色时间不超过5-12分钟，以显微镜下观察为准

2、非特异性背景染色？

原 因：操作过程中冲洗不充分

解决方法：每步冲洗3次每次5分钟

原 因：组织中含过氧化物酶未阻断

解决方法：可再配置新鲜3%H2O2封闭孵育时间延长
