CREB结合蛋白p300抗体背景介绍:Cyclic AMP-regulated gene expression frequently involves a DNA element designated the cAMP-regulated enhancer (CRE). Many transcription factors bind to this element, including the protein CREB which is activated as a result of phosphorylation by protein kinase A. It has been shown that protein kinase A-mediated CREB phosphorylation results in its binding to a nuclear protein designated CBP (for CREB-binding protein). These findings suggest that CBP has many of the properties expected of a CREB co-activator. Another high molecular weight transcriptional adapter protein, designated p300, is characterized by three cysteine- and histidine-rich regions, of which the most carboxy terminal region specifically binds the adenovirus E1A protein. p300 molecules lacking an intact E1A binding site bypass E1A repression even in the presence of high concentrations of E1A. Sequence analysis of CBP and p300 has revealed substantial homology, arguing that these proteins are members of a conserved family of co-activators.

抗体修复方式
修复液:0.01M 枸橼酸(pH6.0)或者0.05M EDTA(pH8.0)
CREB结合蛋白p300抗体,Anti-Ep300抗体抗原修复,必须注意以下问题:
1、修复液的选择。
0.01M枸橼酸(pH6.0):应用zui多的抗原修复液,适用于大多数的抗原,用该液修复后的抗原表达增强,抗原的定性和定位很理想,结果不错;
0.05M EDTA(pH8.0):应用较多的抗原修复液,修复能力较强,对于保存时间较长的切片或目的蛋白表达较弱的组织有很好的修复效果,需要控制好修复时间,否则容易出现较重的背景;
0.01M TBS(pH7.4):中性修复液,适用于大多数抗原,对于核定位蛋白有较好的修复效果;
*:应用较多的抗原修复液,适用于大多数的抗原,使用温度37℃,对于容易脱片的切片有很好的保护作用。

2.抗原热修复时应选择的温度。
据实验认为温度在92℃-98℃是合适的,尤其在95℃为,这是因为:(1)这种温度未达沸腾,切片不容易脱离载玻片;(2)能够解离和破坏与蛋白交联的甲醛醛键等,处理时可以随意选择和确定抗原修复的作用时间。如果选择高压修复,因为温度较高,时间不宜过长
3.抗原修复时有效温度所需持续时间根据各单位的设备条件以及使用的仪器不同,抗原修复时在有效的温度范围内所持续的时间也不一样。
4.抗原修复液必须遵循自然降温规律,否则效果不好或达不到抗原修复的目的。
5.尽量使用足量的抗原修复液,防止切片干涸。
6.切片必须附贴牢固,否则发生掉片。
