1.实验之前准备好所有的标准品（见备用试剂）。建议所有的标准和样本都做复孔。

2.首先把微孔板固定在支架上，分别设好相应的孔（空白孔（空白对照孔不加样品及酶标试剂，其余各步操作相同）、标准孔、待测样品孔），然后在包被板上，标准品和代测样本孔准确加样50μl

3.加10 μL的*到代测样本孔中。（标准孔不加）
4.每孔加入酶标试剂50μl，混合，混合在这一步是很重要的。封板后置37℃温育1个小时。
5.在温育结束前15分钟内准备显色液。（见备用试剂）
6.用下面的一种方法洗板;
7.手洗：将微孔板中的混合物倒入合适的废物器皿中，然后每孔注满稀释好的洗涤液，再到掉，如此重复4次，然后倒置微孔板，将其拍到吸水纸上或者纸巾上，直到拍干。、
（注：洗板的时候要紧握微孔板的两侧，以确保所有的板条不脱落）
8.机洗：吸干所有的孔，然后用洗板机洗板5次，调整洗板机尽可能洗掉所有的液体，设置填充量在350微升/孔/清洗（范围：350-400微升），洗板后，倒置微孔板，将其拍到吸水纸上或者纸巾上，直到拍干。建议洗板机设置的浸泡时间为10秒，震动时间为5秒。
9.每孔依次加入显色剂A 50μl，显色剂B 50μl，封板后置20-25°C显色15分钟.
10.每孔加终止液50μl混合
11.在30分钟以内读酶标仪在450nm波长的O.D值
