elisa 小知识：
辣根过氧化物酶(HRP)是一种糖蛋白，每个分子含有一个氯化血红素(protonhemin)区作辅基。酶的浓度和纯度常以辅基的含量表示。氯化血红素辅基的zui大吸收峰是403nm，HRP酶蛋白的zui大吸收峰是275nm，所以酶的浓度和纯度计算式是(已知HRP的A(1cm 403nm 1%)＝25，式中1％指HRP百分浓度为100ml含酶蛋白1g，即10mg/ml，所以，酶浓度以 mg/ml 计算是HRP的A(1cm 403nm mg/ml=2.5)HRP纯度(RZ)=A403nm/A275nm纯度RZ(Ｒeinheit Zahl)值越大说明酶内所含杂质越少。高纯度HRP的RZ值在3.0左右，zui高可达3.4。用于ELISA检测的HRP的RZ值要求在3.0以上。
