1.专一性强。抗原与抗体的免疫反应是专一反应，而免疫酶技术以免疫反应为基础，所检测的对象是抗原（或抗体），使用的抗体除标记了酶以外，与普通抗体的免疫反应特性并无多大差别。
2.灵敏度高。由于抗体联结上了酶，因此，借助于酶与底物的显色反应，显示抗原与抗体的结合，大大提高了检测的灵敏性，使检测水平接近放射免疫测定法。
3.样品易保存。经过酶反应显示的有色产物大多比较稳定，因此有利于样品的保存。
4.结果易观察。对检测结果即可用肉眼观察，又可用显微镜观察，也可以用分光光度计进行比色测定，还可用显微镜观察。这是因为某些酶反应产物能使电子密度发生改变，从而引起被检测物的显示。
5.可以定量测定。溶液中的抗原物质，应用酶免吸附技术进行比色测定，依据光密度值的变化，可以定量。预计用细胞分光光度计可对组织内的抗原进行免疫酶技术的定量测定。
6.可以大规模测定样品。如有成千上万份样品需要进行检测，免疫酶技术都能在较短时间内完成。
7.仪器和试剂简单。对免疫没技术来说，不需要荧光显微镜，也不需要测定放射性的特殊仪器，所用仪器及试剂均属一般性仪器与试剂，普通实验室及生产应用单位均易购置。
