1、试剂盒特异性因素 
1）固相载体的选择。ELISA中常用的固相载体有微量滴定板、小珠和小试管三种，以微量滴定板zui为常见[1]。它具有良好的吸附性能，孔底透明度高，空白值低，各板之间、同一板各孔之间性能相近。聚苯乙烯ELISA板由于原料的不同和制作工艺的差别，各产品的质量差异很大。因此，在选择试剂盒同时要对其使用的微孔板型号进行认证，评估。 
2）包被物的纯度。在酶联免疫测定尤其是间接ELISA中，试剂的特异性取决于使用抗原的纯度。目前由于技术条件的限制，包被用抗原或抗体的纯度不可能达到100%，所以有些非特异性显色不可避免，只能尽可能提高纯度，提高特异性。目前使用的包被抗原一般为合成多肽抗原。 
3）包被抗体的效价。具有高亲和力和高特异性的包被抗体，是决定试剂特异性的重要方面。 
4 ）封闭。是ELISA中重要的一步，目的是封闭固相载体表面尚未被所占据的空隙[2]，减少后续步骤中非特异性蛋白的干扰。 
2、操作过程中的问题造成假阳性 
1）加样 
对于间接ELISA标本一般都要进行稀释，如果加样不准就会造成误差，尤其当稀释倍数大时，很小的误差，会导致较大的相对误差，使阴性(或弱阳性)标本呈阳性(或阴性)。加样时应将所加物加在ELISA板孔的底部，避免加在孔壁上部，并注意不可溅出，不可产生气泡。目前，大部分血站都已使用全自动酶免加样系统处理标本，可较好地避免以上误差。 
2）洗涤 
在ELISA中正确的洗涤是保证得到可重复结果的关健一步，应引起操作者重视。无论是手工操作还是机器操作，得出不正确的结果常与不正确的洗涤有关，ELISA就是靠洗涤来达到分离游离和结合的酶标记物的目的。通过洗涤以消除残留在板孔中没能与固体抗原或抗体结合的物质，以及在反应过程中非特异性吸附于固相载体的干扰物质。 
3） 温育 
每种试剂都有其佳反应模式，其中温度和温育时间控制是重要因素。因孵育温度高，反应时间长，会造成整板本底高，阳性率高。温育一般用湿盒或水浴，反应板不宜叠放，以保证各板温度都能迅速平衡，为避免蒸发，板上应加盖。 
4）酶标仪判读 
作为记录测定结果的仪器，酶标仪的性能稳定与否，决定结果的可靠度。首先酶标仪应定期进行保养，对滤光片要定期校正；其次酶标仪波长设置要正确，使用双波长，一个检测波长，一个参比波长，以消除微孔板底部划痕、不平、指印或液面高度差异造成的光干扰。此外，在用酶标仪读数时先擦试微孔板底部并压平板条。由于各种酶标仪性能有所不同。
