MRGPRX4蛋白抗体保存条件:Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.
MRGPRX4蛋白抗体,Anti-MRGPRX4抗体溶解方法:
方法1:我们的抗体(冻干粉)已经包含了0.01M PBS、1% BSA和0.1% 防腐剂(叠氮钠或*),您只需要加入灭菌的双蒸水就行了。抗体溶解后,置于-20℃保存,分装后使用,避免反复冻融,可保证至少1年有效;
方法2:也可以只加入一半体积的双蒸水,再用一半体积的甘油补足,置于-20℃保存,这样抗体不会冻,有利于抗体的稳定。
后续试验时,再使用对应的缓冲液稀释成工作液,即用即配。
我们的抗体(冻干粉),在常温放置下,至少一个月有效;若在-20℃下保存(*),至少三年有效。

免疫组化操作步骤:
(一)、仪器设备
1、 18cm不锈钢高压锅或电炉或用微波炉
2、 水浴锅
(二)、试 剂
1、 PBS缓冲液(pH7.2―7.4)
2、 0.01mol/L柠檬酸钠缓冲液(CB,pH6.0,1000ml)
3、 3%甲醇―H2O2溶液:用30% H2O2 和80%甲醇溶液配制
4、 风裱剂:
a. 甘油和0.5mmol/L碳酸盐缓冲液(pH9.0–9.5)等量混合
b. 油和TBS(PBS)配制
5、 TBS/PBS pH9.0–9.5,适用于荧光纤维镜标本;pH7.0-7.4适合光学纤维标本
(三)、操作流程
1、 脱蜡和水化:脱蜡前应将组织芯片在室温中放置60分钟或60℃恒温箱中烘烤20分钟
a. 组织芯片置于二甲苯中浸泡10分钟,更换二甲苯后在浸泡10分钟
b. 无水乙醇中浸泡五分钟
c. 95%乙醇中浸泡五分钟
d. 75%乙醇中浸泡五分钟
2、 抗原修复:用于福尔马林固定的石蜡包埋组织芯片

背景介绍:Mas-related G protein-coupled receptor member X1 (MRGX) is a sensory neuron-specific G protein-coupled receptor that is involved in the function of nociceptive neurons. This integral membrane protein may also regulate nociceptor development and/or the sensation or modulation of pain. There are four members (MRGX1-4) in the human MRGX familly. MRGX1 and MRGX2 receptors stimulate both G?q- and G?i -regulated pathways, while MRGX3 and MRGX4 receptors mainly activate G?q-regulated pathways. G?q proteins are involved in the calcium-signaling pathway downstream of the MRGX receptors. MRGX receptors are unique in that they are expressed in a subset of small dorsal root and trigeminal sensory neurons.
