HLA Class II DR4抗体保存条件:Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

一、抗原抗体反应特点
1.高度特异性,特异性是抗原抗体反应的zui主要特征,特定抗原决定簇只与特定抗体结合;
2.可逆性:抗原与抗体结合形成抗原抗体复合物的过程是一种动态平衡, 其反应式为:Ag+AbAg·Ab;
3.适当浓度和比例;
4.敏感性:在测定血清中某一物质的含量时,化学比色法的敏感度为mg/ml水平,酶反应测定法的敏感度约为5~10μg/ml,免疫测定中凝胶扩散法的敏感度与酶反应法相仿。
二、影响抗原抗体反应的因素
1.电解质 :抗原抗体特异结合后,胶体粒便可发生脱水作用,此时易受电解质影响,如有适当浓度的电解质存在就会使它们失去部分负电荷而相互凝聚,于是出现明显的凝集或沉淀现象。
2.酸碱度:合适的pH为6-8。
3.温度:37℃通常是抗原体反应zui适温度。

免疫染色:
下面以石蜡切片的免疫组化为例演示详细操作流程(SP法)
1.60℃烤片2小时
2.脱蜡。二甲苯脱蜡15分钟,三次
3.水化。依次经过无水乙醇、95%、90%、80%、70%,各5分钟,蒸馏水(或自来水)5分钟
4.PBS(洗液C-0079)洗5分钟,三次
5.抗原修复。枸橼酸缓冲液(修复液,C-0070)(约92-95℃)煮沸(沸水浴)修复15分钟,自然凉至室温。PBS洗5分钟,三次
6.3%双氧水(包含在SP-0023中)消化内源性的过氧化物酶,20分钟,PBS洗5分钟,三次
7.正常山羊血清(包含在SP-0023中)封闭,37℃20分钟
8.一抗孵育,4℃过夜. (抗体稀释液C-0007)
9.PBS洗5分钟,三次
10.二抗孵育,37℃20分钟(二抗为*标记的羊抗兔IgG,包含在SP-0023中),PBS洗5分钟,三次
11.三抗孵育,37℃20分钟(三抗为辣根酶标记的链霉亲和素,包含在SP-0023中),PBS洗5分钟,三次
12.DAB(显色剂C-0010)显色,镜下观察结果,适时终止
13.苏木素(C-0021)复染5分钟,自来水冲洗片刻,1%的盐酸酒精(75%)溶液(此试剂无法销售,请自行配置,配制比例为:100ml 75%酒精+1ml浓盐酸)分化30秒,自来水冲洗5分钟蓝化
14.脱水。依次经过70%、80%、90%、95%、无水乙醇,各5分钟,二甲苯15分钟,三次
15.封片,中性树胶(C-0073)封片

背景介绍:HLA-DRB1 belongs to the HLA class II beta chain paralogs. The class II molecule is a heterodimer consisting of an alpha (DRA) and a beta chain (DRB), both anchored in the membrane. It plays a central role in the immune system by presenting peptides derived from extracellular proteins. Class II molecules are expressed in antigen presenting cells (APC: B lymphocytes, dendritic cells, macrophages). The beta chain is approximay 26-28 kDa. It is encoded by 6 exons. Exon one encodes the leader peptide; exons 2 and 3 encode the two extracellular domains; exon 4 encodes the transmembrane domain; and exon 5 encodes the cytoplasmic tail. Within the DR molecule the beta chain contains all the polymorphisms specifying the peptide binding specificities. Hundreds of DRB1 alleles have been described and typing for these polymorphisms is routinely done for bone marrow and kidney transplantation. DRB1 is expressed at a level five times higher than its paralogs DRB3, DRB4 and DRB5. DRB1 is present in all individuals. Allelic variants of DRB1 are linked with either none or one of the genes DRB3, DRB4 and DRB5. There are 4 related pseudogenes: DRB2, DRB6, DRB7, DRB8 and DRB9.
